【預(yù)期用途】

表皮細(xì)胞生長因子受體（EGFR）是人表皮生長因子受體（HER）家族成員，屬于受體酪氨酸激酶（TKI）家族，為原癌基因c-erbB1表達(dá)產(chǎn)物。EGFR主要位于細(xì)胞膜，被EGF等配體激活后發(fā)生二聚化并引發(fā)胞內(nèi)段產(chǎn)生酪氨酸激酶活性，進(jìn)一步激活下游信號轉(zhuǎn)導(dǎo)，細(xì)胞生長、增殖等多種生理過程。

本試劑盒用于惡性腫瘤（肺癌、結(jié)直腸癌、胰腺癌、鼻咽癌等實(shí)體瘤）患者組織樣本中的Kras基因12和13密碼子突變的定性檢測，為個(gè)體化用藥治療提供依據(jù)。Kras基因突變與惡性腫瘤患者EGFR信號傳導(dǎo)通路的靶向藥物原發(fā)性耐藥有關(guān)。Kras基因野生型的患者能從愛必妥 (Erbitux，又稱西妥昔單抗/Cetuximab)或維克替比(Vectibix，又稱帕尼單抗/Panitumumab)、易瑞沙或特羅凱的治療中獲益，而KRAS基因突變的惡性腫瘤患者無明顯受益于此類靶向藥物治療。

本試劑盒可檢測出如下KRAS基因7種突變：

12密碼子的GGT>AGT、GGT>CGT、GGT>TGT、GGT>GAT、GGT>GCT、GGT>GTT  6種突變和13密碼子的GGC>GAC。
